Commercial Manufacturing at NextPharma Waltrop, Ploërmel and Edinburgh
Hormones are potent, sensitising compounds. Trace carry-over into other products poses a direct patient-safety risk, which is why EU GMP and FDA requirements mandate dedicated production areas, separate equipment and validated cross-contamination controls for hormonal manufacturing. NextPharma addresses this with dedicated hormone production and packaging areas — separate suites, equipment and controls — rather than shared multi-product lines. NextPharma’s Waltrop location covers hormonal solids and semi-solids in fully segregated areas; NextPharma Ploërmel extends that containment to lipid-based softgels across the OEB1 to OEB5 range, and NextPharma Edinburgh adds liquid-filled hard capsule capability in two dedicated high-containment suites. The combination of hormonal solid, semi-solid, softgel and liquid-filled hard capsule capability within a single CDMO network is a unique set of capabilities that NextPharma offers.
Dosage Forms and Technologies Covered
Across the three sites, the following hormonal forms are in routine commercial production:
-
Solids: coated and uncoated tablets
-
Semi-solids: creams, gels, ointments and suppositories
-
Liquids: solutions, suspensions and emulsions
-
Softgel capsules: lipid-based fills, hormone-dedicated production unit
-
Liquid-filled hard capsules (LFHC): lipid-based and semi-solid fills in a two-piece hard-capsule shell
NextPharma Waltrop: Hormonal Solids, Liquids and Semi-Solids
Hormonal bulk production, primary and secondary packaging at NextPharma Waltrop run in segregated hormone areas, physically separated from non-hormonal production on the same site. Ex-proof processing areas, online serialisation and tamper-evidence are standard across all formats.
Dosage forms in routine hormonal production at NextPharma Waltrop:
-
Solids: coated and uncoated tablets; PVC/PVDC/Aclar blisters, flow packs and folding boxes
-
Semi-solids: creams, gels, ointments and suppositories; aluminium tubes, metered-dose pumps and stick packs for hormonal products
-
Liquids: solutions, suspensions and emulsions in glass or plastic bottles and pump sprays
Development services available on-site cover analytics, stability, validation and clinical trial supply manufacturing and packaging for both hormonal and non-hormonal products, including paediatric formulations.
NextPharma Ploërmel: Hormonal Softgels and Liquid-Filled Hard Capsules
Where a hormonal product requires lipid-based delivery, Ploërmel provides a dedicated production unit for hormone-containing softgels and liquid-filled hard capsules (LFHC). The site has over 30 years of experience in softgel technology and runs high-potent handling across the OEB1 to OEB5 range via isolator technologies and engineering controls.
Lipid-based formats are particularly suited to hormonal APIs with bioavailability challenges: the fill matrix supports BCS class II and IV compounds, provides oxidation protection for sensitive molecules, and delivers accurate content uniformity at low doses. Formulation development, scale-up and commercial encapsulation are all available on site.
NextPharma Edinburgh: Hormonal Liquid-Filled Hard Capsules
NextPharma Edinburgh extends hormonal capability to liquid-filled hard capsules (LFHC) with banding as sealing technology. The site runs two dedicated high-containment manufacturing suites for high-potent compounds across the OEB1 to OEB5 range, including hormones, with over 35 years of LFHC experience in an MHRA-licensed and FDA-registered facility.
Lipid-based delivery in a two-piece shell suits hormonal APIs requiring low-dose content uniformity: the API is dosed as a homogeneous liquid dispersion rather than a dry powder blend, which supports accurate dosing and suppresses dust generation at the point of handling. Formulation development, analytical development, clinical manufacture, scale-up and commercial supply are all available on the same campus.
From Feasibility to Commercial Supply
Hormonal programmes follow the same defined sequence at each site, within the applicable containment:
-
Pre-formulation: API characterisation, potency classification and excipient compatibility
-
Analytical development: method development and validation
-
Formulation and galenic optimisation, including paediatric formulations
-
Process development and scale-up under containment
-
Clinical supplies: clinical trial supply manufacturing and packaging under GMP
-
Industrialisation: registration batches, validation, stability and CTD Module 3 registration services
Hormonal products must remain inside contained areas at every step, from bulk manufacture through to packaging. That makes a mid-programme transfer more demanding than for a non-potent product: the receiving site has to re-qualify its containment, re-validate its cleaning procedures and demonstrate equivalent cross-contamination controls. At NextPharma Waltrop, NextPharma Ploërmel and NextPharma Edinburgh, formulation development, scale-up and commercial manufacture all take place within the site’s own contained areas. Hence, the containment strategy and process parameters established during development carry into commercial batches.
One Network Across Every Hormonal Format
From tablets, creams and ointments at Waltrop to lipid-based softgels and high-potent capsules at Ploërmel and liquid-filled hard capsules at Edinburgh, hormonal programmes stay within a single NextPharma network, with matched containment, in-network serialisation and QP release at every step. Waltrop and Ploërmel sit inside the EU regulatory perimeter; Edinburgh operates under MHRA licence in the UK.
